New Environment Sensitive Fluorophores with Color-Tailored Emission: In Vivo Monitoring of Carbonic Anhydrases Expression on Growth of Larval Zebrafish

Protein expression in space and time is significant for understanding important biological processes. This study monitors dynamic progress of newly developed background-free, target responsive strategy; a fluorophore 2,3-dihydroquinolin-4-imine (DQI) that can instantly respond to environmental changes with fluorescence enhancement, revealing a comprehensive platform for in vivo fluorescence bioimaging of target proteins. The density functional theory (DFT) calculation rationalized this response showing considerable change of a dipolar moment between excited and ground states. The novel environment-sensitive property makes DQI an ideal fluorescent turn-on biosensor for continuous and selective biomolecular recognition. An arylsulfonamide-conjugated DQI (SA-DQI) molecule successfully detected human carbonic anhydrase II (hCAII) and SA-DQI can recognize membrane-bound hCAII on HeLa cells without removal of unbound SA-DQI. This chemical strategy allows further instantaneous monitoring of carbonic anhydrase (CA) expression responding to the formation of otolith during zebrafish embryogenesis a promising advancement for study of protein dynamic expression upon chemical and biological stimuli.
